Skupina za eksperimentalno botaniko je del programske skupine Biologija rastlin.
V skupini za eksperimentalno botaniko delamo:
Kaj vse delamo:
- merimo velikost genoma rastlin in endoreduplikacijo (mikrodenzitometrija)
- merimo razvoj rastlinskih organov (povezava med velikostjo genoma, velikostjo celic in ekspresijo encimov)
- spremljamo vpliv težkih kovin na metabolizem rastlin in ekologijo rastlin
- merimo poškodbe DNA zaradi vpliva genotoksičnih snovi (kometni test)
- spremljamo potek programirane celične smrti v razvoju koruznega zrna
- z analizo slike merimo taksonomske znake pri rastlinah
- molekularna analiza linusitina, proteina iz lanu, povezanega s patogenezo
Mikroskop je naš glavni merilni inštrument. Opremljen je takole:
- objektivi 2.5x, 10x, 40x, 63x (oljni), 100x (oljni)
- barvna video kamera (8-bitna, ločljivost 768x576)
- barvna digitalna kamera (12-bitna, ločljivost 1300x1030)
- presevna osvetlitev: 100W halogenska žarnica; svetlo polje, temno polje, fazni kontrast
- fluorescenca: 50W živosrebrna žarnica; ekscitacijski filtri UV (01 - DAPI), moder (09 - FITC) in zelen (15 - Rodamin), vsi longpass emisija, proizvajalec Carl Zeiss
- programsko okolje za zajem in analizo slike (KS400, Carl Zeiss Vision)
- program za zajem in osnovno analizo slike (AxioVision, Carl Zeiss Vision)

Analiza slike je naša glavna metoda:
- mikrodenzitometrija - kvantitativno merjenje obarvanosti jeder s Schiffovim reagentom, rezultat je relativna količina DNA v jedrih
- 3D rekonstrukcija rastlinskih organov (endosperm in koreninska čepica koruze)
- na področju analize slike sodelujemo tudi z drugimi skupinami na Oddelku za biologijo (zoologija, ekologija rastlin, biokemija)
